Whats the difference in the modules? A - B - C - I?
Is it a theme difference, difficulty setting, higher/lower magic?

It is easier to explain if you read the synopsis of each adventure series in the official TLG store.

Checked them out, but I can see no real common thing that links them.
*Some say: Town, Dungeon, Overland, but then there are ones that say straight up dungeon in them as well. This is multiple categories.
*The categories run the breath of levels
Hmm, are they just separated for separate's sake?
Anyone else take a stab? If I were to ask you the difference from an 'S' module and a 'U' what would you say? How about your favorite line and why?

See

for an explanation of the module lettering.
As far as milieu; almost all of the modules from TLG employ a broad sandbox approach, and often employ one or more "dungeons". Heart of Glass is mostly a city adventure, and Wicked Cauldron and Dark Journeys are mostly dungeon based.
